Molecular methods are applied to achieve the distinction of the autochthonous grapevine varieties. Genomic DNA is extracted from fresh leaf tissue and then is used as a template in Polymerase Chain Reactions (PCRs). Many microsatellites (Simple Sequence Repeats –SSRs), including the nine OIV(OrganisationInternationale de la Vigneet du Vin) molecular descriptors, are used to reveal the genetic polymorphism among the various genotypes.
Intra-varietal differences could be traced enabling clonal distinction. However, halting genetic erosion, and conserving and maintaining the autochthonous genotypes, is a primary goal.
